How much bacteriostatic water to add: reconstitution charts by peptide

Most labs add 2 mL of bacteriostatic water to a 5 to 10 mg peptide vial and 3 mL to a 20 mg or larger vial. The concentration is simply mg in the vial / mL of water, and on a U-100 insulin syringe 100 units = 1 mL.

Before you reconstitute
- Let the vial reach room temperature, swab the stoppers, and add the water slowly down the vial wall.
- Swirl gently; never shake. Label the vial with the date and concentration.

Reconstitution FAQ
How much bacteriostatic water should I add to a peptide vial?
A common default is 2 mL for 5 to 10 mg vials and 3 mL for 20 mg and larger vials (most peptide vials hold about 3 mL). The volume is a choice: any volume works as long as the resulting concentration is recorded on the label.
How do I calculate peptide concentration after reconstitution?
Divide the amount in the vial by the volume of water added. 10 mg in 2 mL is 5 mg/mL; 5 mg in 2 mL is 2.5 mg/mL.
How many units are in 1 mL on an insulin syringe?
A U-100 insulin syringe has 100 units per mL, so 10 units is 0.1 mL and 50 units is 0.5 mL.
How do I convert an amount to syringe units?
Units = amount / concentration x 100. For example, 1 mg from a 5 mg/mL solution is 0.2 mL, which is 20 units.
Bacteriostatic water or sterile water?
Bacteriostatic water contains 0.9% benzyl alcohol, which limits microbial growth across repeated punctures of a multi-use vial. Sterile water has no preservative and suits single-use preparations.
