Peptide Reconstitution: Canadian Researcher's Reference Guide

Published 2024-04-15 · Lumera Labs Editorial · Kelowna, BC

Short answer: Add bacteriostatic water (0.9% benzyl alcohol) to the lyophilized peptide vial slowly down the side of the vial wall. Do not shake — gently swirl. Store reconstituted solution at 2–8°C and use within 28 days.

Materials

  • Lyophilized peptide vial (Lumera Labs)
  • Bacteriostatic water (BAC water) — 3 mL or 10 mL
  • Sterile syringe with appropriate gauge needle (typically 25-27g, 1 mL or 3 mL syringe)
  • Alcohol prep pads
  • Sharps container for needle disposal

Bacteriostatic water vs sterile water

Bacteriostatic water (0.9% benzyl alcohol preservative) is preferred for research applications requiring storage of reconstituted solution beyond 24 hours. The benzyl alcohol prevents bacterial growth, extending usable lifespan to ~28 days at 2–8°C.

Sterile water contains no preservative. Use only for single-use applications where the entire reconstituted volume will be consumed within 24 hours, or for compounds incompatible with benzyl alcohol (rare; check compound spec).

Both are available at Lumera Labs accessories.

Volume calculations

The volume of water you add determines the concentration. Common targets:

PeptideVialBAC WaterConcentration
BPC-1575 mg2 mL2.5 mg/mL
TB-50010 mg2 mL5 mg/mL
Semaglutide5 mg2 mL2.5 mg/mL
Tirzepatide10 mg2 mL5 mg/mL
NAD+100 mg5 mL20 mg/mL

Reconstitution technique

  1. Inspect the lyophilized vial — the powder should be a uniform white plug at the bottom or visible as fine flakes against the vial wall.
  2. Wipe the rubber stopper of both vials with an alcohol prep pad and let air-dry.
  3. Draw the calculated volume of bacteriostatic water into your syringe.
  4. Insert the needle into the peptide vial at a 45° angle, against the inner glass wall — not directly onto the powder.
  5. Inject the water slowly, letting it run down the wall. Do not push aggressively.
  6. Remove the needle. Do not shake. Gently swirl the vial in your fingers for 10–15 seconds.
  7. Allow 5–10 minutes for full dissolution. The solution should be clear and free of particulates.

Storage after reconstitution

  • Bacteriostatic water reconstitution: 2–8°C (refrigerator), use within 28 days
  • Sterile water reconstitution: 2–8°C, use within 24 hours
  • Long-term: some peptides can be aliquoted and frozen at -20°C for up to 60 days; this depends on the specific peptide's stability profile (consult your COA or contact us)
  • Avoid: repeated freeze-thaw cycles — degrades peptide structure

Common issues

  • Cloudy solution: incomplete dissolution. Wait 10 more minutes; gently swirl. If still cloudy, peptide may be partially degraded.
  • Particulates floating: stop and contact lumeralabs@proton.me with photos. Possible contamination or degradation.
  • Vacuum lost in vial: normal after reconstitution. Pressure equalizes when water is injected.
  • Powder won't dissolve: some peptides require slight acidification (e.g., 1% acetic acid) before adding BAC water. Check compound-specific spec.

Frequently Asked Questions

What is bacteriostatic water?

Sterile water with 0.9% benzyl alcohol added as a preservative. Used to reconstitute lyophilized research peptides for laboratory storage up to 28 days at 2-8 C.

Can I use tap water or distilled water?

No. Tap water contains microorganisms, and distilled water lacks bacteriostatic preservative. Always use pharmaceutical-grade bacteriostatic or sterile water.

How long does reconstituted peptide last?

With bacteriostatic water at 2-8 C, typically 28 days. With sterile water, 24 hours. Frozen at -20 C in single-use aliquots, up to 60 days for stable peptides.

Can I shake the vial to dissolve faster?

No. Shaking introduces foaming and shear stress that can denature peptide structure. Always swirl gently and allow time.

Where can I buy bacteriostatic water in Canada?

Lumera Labs supplies bacteriostatic water in 3 mL and 10 mL vials at lumerapeptide.com/products/bac-water/. Domestic Canadian shipping.
