Kisspeptin-10 research protocols: GPR54 signalling and the HPG axis

Kisspeptin-10 sits at the top of the reproductive endocrine cascade, upstream of GnRH. Its very short circulating half-life and its position in that hierarchy determine how the published literature designs experiments around it.

Kisspeptin-10 is the C-terminal decapeptide fragment of kisspeptin-54, the product of the KISS1 gene. It retains full agonist activity at KISS1R, the G-protein-coupled receptor still widely referred to by its original orphan designation GPR54. Position in the HPG cascade

The hypothalamic-pituitary-gonadal axis runs in a strict hierarchy, and kisspeptin sits above the classical entry point:

  1. Kisspeptin neurons in the arcuate nucleus and the anteroventral periventricular nucleus release kisspeptin.
  2. Kisspeptin binds KISS1R on GnRH neurons, depolarising them and triggering GnRH release.
  3. GnRH reaches the anterior pituitary through the hypophyseal portal system and stimulates gonadotrophs.
  4. The pituitary releases LH and FSH.
  5. LH and FSH act on the gonads, driving steroidogenesis and gametogenesis.

Because kisspeptin acts one level above GnRH, it is the tool of choice for asking whether a given phenotype originates upstream or downstream of GnRH neurons. If kisspeptin administration restores LH pulsatility, the GnRH neurons are intact and the defect is upstream. If it does not, the lesion is at or below the GnRH neuron.

Half-life constrains the experiment

Kisspeptin-10 clears rapidly. Reported circulating half-life for the decapeptide is in the range of a few minutes, substantially shorter than kisspeptin-54, which persists longer. Two design consequences follow directly:

  • Bolus administration produces a sharp, transient LH response. Sampling windows in published protocols are correspondingly tight, often minutes rather than hours.
  • Continuous infusion produces receptor desensitisation. Sustained KISS1R occupancy downregulates the response, an effect documented across multiple models. Intermittent administration preserves responsiveness.

This desensitisation behaviour is itself a research subject, since it mirrors the well-established desensitisation seen with continuous GnRH agonist exposure.
